摘要:
目的 探讨参附汤对异丙肾上腺素(ISO)所致心肌缺血损伤大鼠的作用及apelin蛋白表达的变化对心肌缺血的影响。方法 采用皮下注射ISO方法建立心肌缺血动物模型。灌胃给予大鼠不同剂量的(3、6、12 g·kg-1)参附汤,模型组与空白组灌胃等体积蒸馏水,阳性组灌胃参附注射液6.67 g·kg-1,连续灌胃15 d; 从d 11开始,灌胃1 h后,皮下多点注射ISO,连续5 d造模。造模后考察参附汤对心肌组织形态学、血清中心肌酶含量、心肌组织中apelin mRNA含量及血清中apelin蛋白含量的影响,并探讨心肌酶与apelin表达变化的相关性。结果 参附汤可以明显减轻心肌组织坏死程度; 与模型组相比,参附汤中、高剂量组肌酸激酶(CK)值及各剂量组的乳酸脱氢酶(LDH)值均明显降低(P<0.05或P<0.01); 参附汤中、高剂量组大鼠血清apelin蛋白含量及心肌组织mRNA含量与模型组相比,均明显升高(P<0.05,P<0.01); 此外,血清apelin与心肌酶CK、LDH的表达呈负相关。结论 参附汤具有抗ISO所致大鼠心肌缺血作用,该作用机制与其促进apelin mRNA表达,使apelin蛋白含量增多,从而抑制心肌酶生成,改善心肌缺血有关。
Abstract:
Aim To study the effects of Shenfu soup on the ISO-induced myocardial ischemia injury model of rats and the influence of apelin level changes on myocardial ischemia. Methods The myocardial ischemia injury model of rats was established by the subcutaneous injection of ISO. For 15 days, rats in Shenfu groups were given by gavage 3, 6 and 12 g·kg-1 Shenfu soup and rats in SF injection group were given 6.67 g·kg-1 Shenfu injection, rats in sham group and model group were given the same volume of distilled water. Rats were subcutaneously multi-point injected ISO for 5 days on the eleventh administrating day. The effects of Shenfu soup on myocardial morphology, serum myocardial enzyme levels, apelin mRNA level in serum and apelin protein level in myocardial tissue, and relationship between myocardial enzyme and apelin expression were evaluated. Results Shenfu soup could reduce the degree of myocardial tissue necrosis and compared with model group, CK values and LDH values of rats in middle-and high-dose groups were significantly lower(P<0.05 or P<0.01). However, the apelin mRNA level in myocardial tissue and apelin protein level in serum of the rats in middle- and high-dose groups compared with model group had increased(P<0.05, P<0.01). Meanwhile, serum levels of apelin and the expression of myocardial enzyme CK and LDH were negatively correlated. Conclusion Shenfu soup has a protective effect on myocardial ischemia injury in rats induced by ISO and the mechanism is involved with the promotion of apelin mRNA and protein expression, the inhibition of myocardial enzyme production and the improvement of myocardial ischemia.
