摘要:
目的 研究黄芪建中汤(Huangqi Jianzhong decoction, HQJZ)对大鼠十二指肠溃疡(duodenal ulcer, DU)的治疗效应及其对TLR-2介导的肠黏膜免疫屏障功能的影响。方法 将大鼠分为正常组、模型组、阳性药组和HQJZ组,采用复合病因法构建DU模型。观测大鼠整体状态; 对DU损伤进行评分; HE染色观察十二指肠病理改变; ELISA法检测细胞因子含量; 荧光定量PCR和免疫组化法分别检测mRNA及蛋白表达。结果 与正常组比较,模型大鼠十二指肠黏膜发生明显溃疡病变; 血清IL-4、IL-10含量明显下降,TNF-α含量明显升高; 黏膜TLR-2、MyD88 mRNA及蛋白表达明显上调。给予HQJZ治疗后,大鼠体质量、进食量、肛温明显回升; 溃疡指数明显降低; 十二指肠黏膜形态恢复正常,绒毛高度和隐窝深度明显增加; IL-4、IL-10含量明显增加,TNF-α含量明显下降; 黏膜TLR-2、MyD88 mRNA及蛋白表达明显下调。结论 HQJZ可促进DU愈合,其机制可能与干预TLR-2介导的肠黏膜免疫屏障功能有关。
Abstract:
Aim To study the therapeutic effect of Huangqi Jianzhong decoction(HQJZ)on duodenal ulcer(DU)in rats and its effect on intestinal mucosal immune barrier function mediated by TLR-2.Methods SD rats were divided into normal group, model group, positive drug group and HQJZ group.DU model was established by methods of multiple factors.The body weight, food intake, and rectal temperature were measured.The ulcer index(UI)was calculated.The pathological changes of duodenal mucosa were observed by HE staining.The contents of cytokines were detected by ELISA.The expression of mRNA and protein were detected by qRT-PCR and immunohistochemistry, respectively.Results Compared with normal group, the body weight, food intake, and rectal temperature of model group decreased significantly.The UI significantly increased.The contents of serum IL-4 and IL-10 markedly decreased, and TNF-α content evidently increased.The expressions of TLR-2 and MyD88 mRNA and protein significantly increased.Compared with model group, after treatment with HQJZ, the body weight, food intake, and rectal temperature were significantly up-regulated, while the UI was significantly down-regulated.The morphology of duodenal mucosa returned to normal status, and the villus height and crypt depth increased significantly.The levels of serum IL-4 and IL-10 were significantly elevated, while TNF-α significantly declined.The expressions of TLR-2 and MyD88 mRNA and protein were apparently down-regulated.Conclusions HQJZ can promote the healing of DU, and the mechanism may be related to the intervention of TLR-2 mediated intestinal mucosal immune barrier function.
